Abstract: A method of generating an image or video of a field of interest of a sample which includes obtaining i) a first RGB image from a field of interest of a sample, and ii) hyperspectral data from a subarea of the field of interest, extracting an RGB image of the subarea from the first RGB image of the field of interest, applying the hyperspectral data of the subarea to conduct a spectroscopic analysis of a sample thereby generating spectral parameters, inputting i) the spectral parameters, and ii) the first RGB image, collectively as training input data to a deep learning model (DLM), training the DLM with the training input data thus generating a trained DLM, obtaining and inputting a second RGB image of about the field of interest to the trained DLM, and outputting from the trained DLM a spectral map for the field of interest.

Abstract: A method of in situ point spread function (PSF) retrieval is disclosed which includes encoding 3D location of molecules into PSFs, receiving molecule-generated images containing PSFs, segmenting the images into sub-PSF, initializing template PSFs from a pupil function, determining a statistical measure of a predetermined function between the sub- and template PSFs, associating each of the sub-PSFs with a template PSF, aligning and averaging the sub-PSF, applying a phase retrieval algorithm to the averaged sub-PSFs to update the pupil function, regenerating the template PSFs, repeating until a difference between a new and a prior generation pupil function is below a predetermined threshold, generating in situ PSFs from the last pupil function, and applying a maximum likelihood estimation algorithm based on the in situ PSFs and the sub-PSF to thereby generate lateral and axial locations of molecules.
Abstract: In some aspects, mutant or variant Fc domains are provided that exhibit increased binding to FcRn and increased half-life after administration in vivo. The Fc domain may be comprised in a glycosylated or aglycosylated antibody. Methods for using the mutant or variant Fc domains or polypeptides comprising the mutant or variant Fc domains are also provided.

Abstract: Emerging evidence indicates that diminished activity of the vasoprotective axis of the renin-angiotensin system, constituting angiotensin converting enzyme2 (ACE2) and its enzymatic product, angiotensin-(1-7) [Ang-(1-7)] contribute to pulmonary hypertension (PH). However, clinical success for long-term delivery of ACE2 or Ang-(1-7) would require stability and ease of administration to increase patient compliance. Chloroplast expression of therapeutic proteins enables their bioencapsulation within plant cells to protect from acids and gastric enzymes; fusion to a transmucosal carrier facilitates effective systemic absorption. Oral feeding of rats with bioencapsulated ACE2 or Ang-(1-7) attenuated monocrotaline (MCT)-induced increase in right ventricular systolic pressure, decreased pulmonary vessel wall thickness and improved right heart function in both prevention and reversal protocols.

Abstract: In shotgun proteomics, generally only a fraction of peptides from a parent protein are actually detected. Because a large portion of the protein sequence is not detected, it is often impossible to determine whether the expressed protein is present in a modified, spliced, or truncated form. Provided herein are methods and systems for analyzing polypeptides which allow for the increase of the mean sequence coverage of a protein concomitant with bioinformatics analysis in order to distinguish putative proteoforms with improved amino acid resolution. Aspects of the invention include (1) a deep sequencing strategy to provide more protein sequence coverage than is typically achieved, and (2) a computational approach to view protein expression across its full length and identify regions of the protein that are potentially subject to such regulation. This technology has global utility in proteomics and will be of particular use for the analysis of biosimilar protein drug therapeutics.
Abstract: Disclosed embodiments include computer-implemented methods and systems that can efficiently generate highly relevant recommended search terms to clinicians. A hybrid collaborative filtering model recommends search terms for a specific patient to the clinician. The model draws on information from patients' clinical encounters and the searches that were performed during the clinical encounters. To generate recommendations, the model uses search terms which are (1) frequently co-occurring with the ICD codes recorded for the patient and (2) highly relevant to the most recent search terms. One variation of the model uses only the most recent ICD codes assigned to the patient. Another variation uses all ICD codes. Comprehensive experiments of embodiments of the methods and systems have demonstrate high levels of performance.

Abstract: The present disclosure provides compositions and methods for decreasing permissiveness to infection by PRRSV. The methods involve inserting peptides or oligopeptides into the CD163 SRCR domain 5 to disrupt the protein structure and thereby prevent PRRSV infection. The deletion of the exon 13 region of PSTII eliminates infection. The replacement of cysteine residues with alanines in SRCR5 as a means to disrupt disulfide bond formation. The final method is the deletion of, or amino acid substitution within the SRCR4-5 interdomain region.
