Abstract: A lytic reagent composition and methods for differential analysis of leukocytes are disclosed. In embodiments, the analysis may utilize optical measurements in flow cytometry based hematology analyzers. The reagent system includes an anionic surfactant in a hypotonic solution, an inorganic buffer to maintain the pH in a range from about 6 to about 10, and optionally a leukocyte stabilizer. The reagent system is used to lyse red blood cells and stabilize the leukocytes to enable multi-part differentiation of leukocytes in a near physiologic pH environment on a flow cytometry based hematology analyzer using axial light loss, light scatter intensity, high-numerical aperture side scatter, and time-of-flight measurements.

Abstract: A device for conducting an assay to determine the presence or amount of an analyte in a fluid sample. The device includes a flow path that facilitates fluidic flow by capillary action. Abstract: The present invention provides compositions and methods for administering florfenicol to mammals. The compositions contain a prodrug of florfenicol in a pharmaceutically acceptable carrier. In one embodiment the prodrug is an esterized form of florfenicol. Examples of suitable prodrugs include one or a combination of one or a combination of the following: florfenicol acetate, florfenicol propionate, florfenicol butyrate, florfenicol pentanoate, florfenicol hexanoate, florfenicol heptanoate, florfenicol octanoate, florfenicol nanoate, florfenicol decanoate, florfenicol undecanoate, florfenicol dodecanoate, and florfenicol phthalate. In another embodiment the prodrug is converted into the florfenicol in vivo by the action of one or more endogenous esterases. The invention also provides new compounds, pharmaceutical compositions containing the compounds, and methods for their administration.

Abstract: The present invention provides compositions and methods for extending the release times and lowering the toxicity of pharmacologically active compounds. The compounds comprise a salt of the pharmacologically active compound with a lipophilic counterion and a pharmaceutically acceptable water soluble solvent combined together to form an injectable composition. The lipophilic counterion may be a saturated or unsaturated C8-C22 fatty acid, and preferably may be a saturated or unsaturated C10-C18 fatty acid. When injected into a mammal, at least a portion of the composition precipitates and releases the active compound over time. Thus, the composition forms a slowly releasing drug depot of the active compound in the mammal. Therefore, the present invention enables one to provide a controlled dose administration of the active compound for a periods of up to 15 days or even longer.

Abstract: The present invention provides devices and methods for determining the presence or absence of pathogens in a biological sample and for the concurrent determination of the antimicrobial susceptibility of pathogens present. The devices and methods of the present invention are able to make the determinations without pre-selection of target bacteria. Instead, all organisms from the sample are applied to the device. In wells where one or more antimicrobial agents are present only pathogens resistant to the antimicrobial agent grow. This allows the user to determine the presence of target pathogens and concurrently determine the resistance patterns of the organisms without the need to subculture the organisms. The present invention also provides methods of performing a business service of performing the determinations described above, and kits containing the devices and descriptions of the methods.

Abstract: A pipette tip includes a main body having an upper end, a lower end and a sidewall extending between the two ends. A bore extends axially through the main body between the upper and lower ends. The lower end has a bottom surface in which is formed a central opening that communicates with the axial bore to allow the passage of fluid therethrough. The bottom surface is formed with at least one groove that may be concentrically disposed or spirally disposed about the central opening. The at least one groove acts to inhibit fluid passing through the central opening from at least partially flowing along the outer surface of the sidewall from the central opening toward the upper end of the main body of the pipette tip.

Abstract: An optical system for an internal drum readout apparatus is disclosed. The optical system includes a hollow cylindrical segment defining a central axis, a support structure configured and adapted to translate along the central axis, a mirror mounted on the support structure for translational movement therewith and for rotational spinning around the central axis, a light source mounted to the support structure for providing a beam capable of being directed along the central axis which in turn is directed against the medium thereby producing a stimulated light, a detector coaxially aligned with the central axis, the detector being configured and adapted to absorb stimulated light direct toward and reflected off of the angled mirror, and a shroud mounted on the support structure for blocking stimulated light not directed toward the angled mirror, wherein the stimulated light not directed toward the angled mirror would otherwise degrade the medium prematurely.

Abstract: A method for determining the presence or amount of a gadolinium chelate in a biological sample. The method includes contacting a biological sample with a dye selected from arsenazo III or chlorophosphonazo at low pH, and measuring the absorbance of the sample, thereby determining the presence or amount of gadolinium in the sample. A method for determining glomerular filtration (GFR) rate in a mammal. The method includes administering to the mammal an amount of a gadolinium chelate and determining the concentration levels of the chelate in biological samples taken from the animal at plurality of intervals following administration of the chelate. The concentration levels of the chelate are correlated to GFR.
Abstract: A method and device for determining a feline immunodeficiency virus infection or vaccination in an animal. The method includes contacting a biological sample from a felid with various FIV polypeptides and determining the binding of antibodies in the sample to the polypeptides. The determination of whether an animal is infected with FIV or has been vaccinated against FIV can be determined by measuring the animal's immune response to an FIV env polypeptide. A device for detecting FIV antibodies is provided.

Abstract: Systems and methods for identifying populations of events in a multi-dimensional data set are described. The populations may, for example, be sets or clusters of data representing different white blood cell components in sample processed by a flow cytometer. The methods use a library consisting of one or more one finite mixture models, each model representing multi-dimensional Gaussian probability distributions with a density function for each population of events expected in the data set. The methods further use an expert knowledge set including one or more data transformations and one or more logical statements. The transformations and logical statements encode a priori expectations as to the populations of events in the data set. The methods further use program code by which a computer may operate on the data, a finite mixture model selected from the library, and the expert knowledge set to thereby identify populations of events in the data set.
