Preferably, such methods and systems combine FRET (Förster Resonance Energy Transfer)-based proximity sensing of labeled nucleotides in or near a DNA polymerase's active site with FRET-based monitoring of the conformational changes in the polymerase that occur during nucleotide incorporation.
