Abstract: Systems and methods for computational analysis of chemical data to predict binding targets of a chemical are provided. A plurality of chemical pairs is established, each including a first chemical for which binding targets are to be predicted and a respective one of the second chemicals. For each chemical pair, values of at least two datatypes of the first chemical can be compared to values of the at least two datatypes of the respective one of the plurality of second chemicals in the chemical pair to generate a similarity score. The similarity scores can be converted to a likelihood value. For each chemical pair, a total likelihood value can be determined based on respective likelihood values for each of the at least two datatypes of the chemical pair. A candidate binding target is predicted to bind to the first chemical, based on the total likelihood value of each chemical pair.

Abstract: Provided are nucleic acid-based sensors, kits that include such sensors, and methods for making and using such sensors. The sensors permit detection of a broad array of target agents, such as nucleic acids (e.g., DNA and RNA), proteins, cells, and small molecules (e.g., toxins and metals). In some examples, the sensors determine the activity of a protein, such as an enzyme's activity. The assays provide easy-to-read signals for point-of-care diagnosis. The sensors control the conformational switch for recognizing targets and to trigger the subsequent assembly of enzyme/cofactor structures for producing and amplifying signals.

Abstract: Clostridium difficile infection is the leading cause of hospital acquired antibiotic-associated diarrhea in the US (Bartlett, in 2006). The increased prevalence of circulating C. difficile strains poses a significant health threat to US health care facilities. Strains expressing the toxin C. difficile Transferase (CDT), in addition to Toxins A and B (TcdA and TcdB), are more virulent and are associated with higher mortality rates (Bacci et al., 2011). We recently identified a protective role for eosinophils against C. difficile pathogenesis (Buonomo et al., 2016). We have also defined CDT's ability to increase host inflammation and suppress protective eosinophils through a TLR2 dependent mechanism (Cowardin et al., 2016). How CDT promotes virulence and eosinophil suppression via TLR2 is still under investigation.
