Abstract: The present disclosure provides for characterization of normal flora and identifying biomarkers in the gut of healthy, neurotypical subjects. Aspect of the disclosure provide for the characterization of the gut microbiome in ADS subjects, characterized by reduced richness and significant loss of the ‘Prevotella-like enterotype’ compared to neurotypical subjects. The relative abundance of genera Prevotella, Coprococcus, Prevotellaceae and Veillonellaceae are significantly lower in autistic children than in neurotypical children. Further, Prevotella, is one of the three main classifiers for the human enterotypes, along with Bacteroides and Ruminococcus. These three core genera are among main contributors in the principle component analysis. ‘Prevotella-like enterotype’ was absent in the autistic group, while neurotypical samples showed an even distribution among the three enterotypes.

Abstract: Provided herein are methods for increasing the yield of an extracellular product synthesized by an organism cultured in a continuous aerobic fermentation system. The extracellular product yield is increased through the use of an organism modified to decreased production of polyhydroxyalkanoate, to increase production of the extracellular product, and to include promoters that can be inducible in response to nutrient limitation conditions. The extracellular product yield is also increased by operating the continuous fermentation system under particular nutrient limitation conditions. Also provided are non-naturally occurring organisms that have been modified for use with the provided methods, and extracellular products made using the provided methods.

Abstract: The present disclosure provides methods for generating sugars from a cellulosic biomass. The methods combine treatment of the biomass using a high-shear milling device and saccharification of the biomass to partially hydrolyze the biomass. The biomass can be saccharified either after or simultaneously with the high-shear milling treatment. The partially hydrolyzed biomass is then separated into a solids stream with saccharification enzymes, and a liquid stream with sugars. The solids stream and associated enzymes are further incubated under saccharification conditions to produce additional sugars, or are recycled and added to fresh biomass, which is saccharified under high-shear milling conditions. The methods result in improved conversion of cellulosic biomass to glucose.

Abstract: Human-derived probiotic strains Lactobacillus paracasei KBL382, Lactobacillus paracasei KBL384 and Lactobacillus paracasei KBL385 are described, as well as uses of the strains for improving intestinal health and treating or preventing intestinal diseases. The strains have excellent anti-inflammatory and immunomodulatory functions, superb strengthening effects on tight junctions of the intestinal tract wall, suppress enteritis-induced weight loss and colon length reduction, thereby exhibiting therapeutic effects for enteritis, and significantly alleviate the symptoms of atopic dermatitis. The strains can be used as probiotic material for enhancing anti-inflammatory effects, strengthening immunity, improving intestinal health functions, and alleviating allergic diseases.
