  • Publication number: 20240209329
Abstract: Some aspects of this disclosure provide a fusion protein comprising a guide nucleotide sequence-programmable DNA binding protein domain (e.g., a nuclease-inactive variant of Cas9 such as dCas9), an optional linker, and a recombinase catalytic domain (e.g., a tyrosine recombinase catalytic domain or a serine recombinase catalytic domain such as a Gin recombinase catalytic domain). This fusion protein can recombine DNA sites containing a minimal recombinase core site flanked by guide RNA-specified sequences. The instant disclosure represents a step toward programmable, scarless genome editing in unmodified cells that is independent of endogenous cellular machinery or cell state.

  • Publication number: 20240212680
Abstract: A device, such as Network Microphone Device or a playback device, receives an indication of a track change associated with a playback queue output by a media playback system. In response, an input detection window is opened for a given time period. During the given time period the device is arranged to receive an input sound data stream representing sound detected by a microphone. The input sound data stream is analyzed for a plurality of command keywords and/or a wake-word for a Voice Assistant Service (VAS) and, based on the analysis, it is determined that the input sound data stream includes voice input data comprising a command keyword or a wake-word for a VAS. In response, the device takes appropriate action such as causing the media playback system to perform a command corresponding to the command keyword or sending at least part of the input sound data stream to the VAS.

  • Publication number: 20240211751
Abstract: Methods, systems, and apparatus, including computer programs encoded on a computer storage medium, for generating an output sequence from an input sequence. One of the methods includes, at each of a plurality of generation time steps: generating a combined sequence for the generation time step that includes the input sequence followed by the output tokens that have already been generated as of the generation time step; processing the combined sequence using a self-attention decoder neural network to generate a time step output that defines a score distribution over a set of possible output tokens; and selecting, using the time step output, an output token from the set of possible output tokens as the next output token in the output sequence.

  • Publication number: 20240213996
Abstract: Embodiments described herein can enable minimum processing performed at a signal device level, such that the majority of the non-domain-specific processing can be offloaded to a processing device. For example, the processing device can receive signal data from a signal device, preprocess the signal data to obtain preprocessed signal data having a data format for domain-specific processing by software executed by at least one processing unit of a processing platform, and provide the preprocessed signal data for domain-specific processing by the software executed by the at least one processing unit.

  • Publication number: 20240207359
Abstract: As disclosed herein, ?-arrestin1 and ?-arrestin2 levels are highly elevated in brains of FTLD-tau patients suggesting that both ?-arrestin1 and ?-arrestin2 are elevated in the brains of patients with AD and FLTD. The current work also shows that when ?-arrestin2 is overexpressed, tau levels become elevated. The data indicate that ?-arrestin2 reduces tau clearance by impairing p62-mediated autophagy, a role carried out by the oligomerized form of ?-arrestin2. Therefore, disclosed herein are ?-arrestin oligomerization inhibitors that can be used to prevent ?-arrestin oligomerization and therefore the accumulation of tau in cells, i.e. tauopathy. Also disclosed are methods of treating a tauopathy in a subject that involve administering to the subject a therapeutically effective amount of a ?-arrestin oligomerization inhibitor disclosed herein.

  • Publication number: 20240211571
Abstract: Methods, systems, and apparatuses are described herein for improving computer authentication processes using vocal confidence processing. A request for access to an account may be received. An authentication question may be provided to a user. Voice data indicating one or more vocal utterances by the user in response to the authentication question may be received. The voice data may be processed, and a first confidence score that indicates a degree of confidence of the user when answering the authentication question may be determined. An overall confidence score may be modified based on the first confidence score. Based on determining that the overall confidence score satisfies a threshold, data preventing the authentication question from being used in future authentication processes may be stored. The data may be removed when a time period expires.

  • Publication number: 20240209377
Abstract: The invention provides non-opioid pain therapeutic compositions that include an antisense oligonucleotide (ASO) complementary to an identified target on a NaV channel mRNA. The ASO hybridizes to its target RNA and forms a duplex that recruits RNase H to degrade the RNA, thereby downregulating NaV channel synthesis, which inhibits the neuron's ability to contribute to the perception of pain. The ASO targets one of the specific identified targets, and may be provided as a gapmer that includes a central DNA segment flanked by modified RNA wings. When the composition is delivered to dorsal root ganglion (DRG) neurons in vitro the DRG neurons exhibit a dose-dependent knockdown of NaV 1.7 NaV 1.8 or NaV 1.9.

  • Publication number: 20240214305
Abstract: Aspects of the present disclosure involve systems, methods, computer program products, and the like, for utilizing a CIC value field in signaling information of a communication to provide an identification of the ingress network to an egress or receiving network of a long distance telecommunications network. The system and method provides for the provisioning of a signaling CIC for an ingress trunkgroup or network to a telecommunications network for downstream signaling purposes by overriding a received CIC value with a provisioned CIC value specific to the ingress network. This provisioned CIC value identifies the ingress network to the long distance network to the egress network for use by the egress network.

  • Publication number: 20240208985
Abstract: Herein is reported a class of chiral compounds with paradoxical effects on the androgen receptor (AR). The (R)-enantiomers behave like classical anti-androgens while the (S)-enantiomers activate AR signaling. In castration-resistant prostate cancer, the change during the course of therapy to growth in the presence of AR targeted therapeutics, a harbinger of progression to lethal disease, is commonly attributed to acquired mutations of the AR-ligand binding domain. This is the first report of an antagonist -agonist duality solely due to structural enantiomerism, without any modification to the AR binding site.
