  • Publication number: 20250135039
Abstract: The present disclosure includes MGMT modifications that cause MGMT resistance to MGMT inhibitors. The present disclosure also includes in vivo, in vitro, and ex vivo modification of MGMT-encoding nucleic acids to encode and express an MGMT variant that is resistant to MGMT inhibitors. Inhibitor-resistant MGMT modifications and modification of an MGMT-encoding nucleic can occur or be used in conjunction with a therapeutic modification, e.g., in a single cell. Expression of inhibitor-resistant MGMT can selectively protect modified cells from a selection regimen, such as a selection regimen including an MGMT inhibitor and an alkylating agent. Accordingly, in vivo, in vitro, and ex vivo modification of MGMT-encoding nucleic acids can be used in gene therapy to select for modified cells.

  • Patent number: 12288389
Abstract: A method for learning disentangled representations of videos is presented. The method includes feeding each frame of video data into an encoder to produce a sequence of visual features, passing the sequence of visual features through a deep convolutional network to obtain a posterior of a dynamic latent variable and a posterior of a static latent variable, sampling static and dynamic representations from the posterior of the static latent variable and the posterior of the dynamic latent variable, respectively, concatenating the static and dynamic representations to be fed into a decoder to generate reconstructed sequences, and applying three regularizers to the dynamic and static latent variables to trigger representation disentanglement. To facilitate the disentangled sequential representation learning, orthogonal factorization in generative adversarial network (GAN) latent space is leveraged to pre-train a generator as a decoder in the method.

  • Publication number: 20250120647
Abstract: Systems, methods, and computer programs for monitoring the disease progression of a Parkinson's patient are provided. Specifications for a Parkinson's patient may defined in a computer system by a physician. The specifications may contain threshold values. At repeating points in time, a message may including a request to perform a mobility test may be output by the computer system to the patient. Mobility parameters may be determined as a result of the performance of the mobility test. The mobility parameters may be compared with the threshold values to determine one or more deviations between the one or more mobility parameters and the one or more threshold values. The mobility parameters, the deviations, or a combination thereof may be made available to the physician by outputting or storing the mobility parameters or the deviations or by transmitting the mobility parameters or the deviations to a separate computer system.
