  • Publication number: 20220288001
Abstract: As disclosed herein, we have discovered that creatine is a critical molecule buffering ATP levels in cancer-targeting CD8 T cells through maintaining a readily available high-energy phosphate reservoir. Building upon this discovery, we have designed a number of methods for modulating energy metabolism in a population of tumor-infiltrating CD8 T cells, methods that can be adapted for use in therapeutic regimens for the treatment of cancer. Illustrative embodiments of the invention include methods for enhancing tumor-infiltrating CD8 T cells ability to mount and sustain a response to tumor cells comprising increasing the concentrations of creatine available for tumor-infiltrating CD8 T cells energy metabolism, thereby enhancing the ability of the tumor-infiltrating CD8 T cells to mount and sustain a response to the tumor cells.

  • Publication number: 20220211757
Abstract: Aspects of the present disclosure relate to methods and compositions related to the preparation of immune cells, including engineered T cells comprising at least one exogenous ?? T cell receptor, for example one that is selected to target a specific disease or pathogen (e.g., cancer or COVID-19). The T cells may be produced from human hematopoietic stem/progenitor cells and are suitable for allogeneic cellular therapy because they do not induce graft-versus-host disease (GvHD) and resist host immune allorejection. Consequently, such cells are suitable for off-the-shelf use in clinical therapy.

  • Publication number: 20210324035
Abstract: Tumor-specific T cell receptor (TCR) gene transfer enables specific and potent immune targeting of tumor antigens. The canonical cancer-testis antigen, NY-ESO-1, is not expressed in normal tissues but is aberrantly expressed across a broad array of cancer types. It has also been targeted with A2-restricted TCR gene therapy without adverse events or notable side effects. To enable the targeting of NY-ESO-1 in a broader array of HLA haplotypes, we isolated TCRs specific for NY-ESO-1 epitopes presented by four MHC molecules: HLA-A2, -B07, -B18, and -C03. Using these TCRs, we have developed an approach to extend TCR gene therapies targeting NY-ESO-1 to patient populations beyond those expressing HLA-A2.

  • Patent number: 11123033
Abstract: Disclosed is a molecular imaging method for evaluating liver regeneration ability after ALPPS. The method includes steps of: 1) preparing a VX2 rabbit liver cancer model; 2) performing ALPPS for the VX2 rabbit liver cancer model; 3) synthesizing 18F-Fluoromethylcholine (18F-Methylcholine, 18F-FCH); 4) PET/CT imaging and data processing for 18F-FCH. The disclosure is the first to propose the use of 18F-FCH PET/CT to monitor the proliferative capacity of residual liver, and further indirectly reflect the increased ability of cell membrane synthesis on the basis that 18F-FCH has higher choline metabolism in residual liver tissue, so that the liver regeneration ability after ALPPS is evaluated by the molecular imaging method, thereby providing important new ideas for the clinical selection of ALPPS to choose the best time for second-stage surgery.
