Publication number: 20200354669Abstract: Embodiments described herein generally provide for the scheduled feeding of cells in a cell expansion system. A schedule to proactively and automatically increase and/or maintain inlet rates of media to feed cells may be created, in which inlet rates to the intracapillary portion (or extracapillary portion) of a bioreactor may be increased or maintained according to the schedule. Such schedule may be conservative or aggressive or a combination thereof, for example. Multiple schedules may be used. Scheduled media exchanges may also be included. By following a feed schedule, the monitoring of metabolite levels may be optional. Inlet rates may be increased or maintained without manual manipulation. Media usage may also be more predictable. In an embodiment, a custom task(s) may be created to follow a desired feed schedule. In another embodiment, a pre-programmed task(s) may be used for the scheduled feeding of cells.Type: ApplicationFiled: July 28, 2020Publication date: November 12, 2020Applicant: Terumo BCT, Inc.Inventor: Boah VANG

Publication number: 20200354670Abstract: The present disclosure relates to a mutant algal strain, wherein said mutant algal strain shows upregulation of mRNA transcripts encoding urea carboxylase, ?-15-?3-desaturase and down-regulation of mRNA transcripts of gene encoding triacylglycerol lipase. Said mutant algal strain of the present disclosure is tolerant to low temperature and thus can be grown over a wide temperature range. The strain shows enhanced biomass and fatty acid production and enhanced growth rate and nitrogen metabolism over a wide temperature range of about 10° C. to about 37° C., wherein said enhancement is in comparison to the wild type algal strain. The present disclosure further provides a method of obtaining said mutant algal strain and method of producing industrially relevant products such as fatty acids from said mutant algal strain.Type: ApplicationFiled: March 30, 2020Publication date: November 12, 2020Inventors: Chitranshu Kumar, Bhaskar Bhadra, Niraja Soni, Saranya Karuppasamy, Shradha Khater, Venkata Subhash G, Aniket A. Publication number: 20200354671Abstract: The invention pertains to methods and apparatuses for rapid production of organized tissue precursors with different sizes and geometry. An embodiment of the invention provides a method of producing 3D tissue structures, the method comprising: producing a plurality of patterned monolayers of cells and sequentially overlaying the plurality of patterned monolayers of cells to produce a stack of monolayers of cells. The patterns of cells in the plurality of monolayers can be designed in a manner which produces a tissue or a portion of a tissue when the plurality of monolayers is overlaid in to a stack. The invention also pertains to an apparatus for producing a 3D tissue module. The apparatus comprises: a three-dimensional positioning system, a contact stamp, a contact stamp storage container, a contact stamp holder, a contact stamp exchange unit, and a contact stamp actuator.Type: ApplicationFiled: October 6, 2016Publication date: November 12, 2020Applicant: UNIVERSITY OF SOUTH FLORIDAInventors: MICHAEL C. Publication number: 20200354674Abstract: Embodiments of a method of modulating a potency level phenotype of a source population of cells include culturing the source cell population in a liquid medium within a cell culture incubator, the incubator is configured to independently regulate one or more variable parameters of gaseous conditions within the incubator, the more than one variable atmospheric parameters within the incubator being regulated independently of each other and independently of ambient external gaseous conditions. The method further includes regulating the one or more variable parameters of the gaseous condition within the incubator such that at least one of the one or more variable parameter differs from an ambient level of the respective variable parameter, and as a consequence of such difference from ambient conditions, the subset population is driven from a first potency level phenotype to a second potency level phenotype.Type: ApplicationFiled: December 18, 2018Publication date: November 12, 2020Inventors: James LIM, Bruce ADAMS, Zack PAPPALARDO, Brian FETH
