Abstract: The present invention uncovers that mutations in GALNT3 gene encoding UDP-N-acetyl-alpha-D-galactosamine:polypeptide N-acetylgalactosaminyltransferase (GalNAc-T3) cause familial tumoral calcinosis (FTC). Methods and pharmaceutical compositions useful for treating disorders associated with abnormal phosphate metabolism are provided. Specifically, inducers of GalNAc-T3 can be used to treat hyperphosphatemia related disorders such as FTC, and on the other hand, inhibitors of GalNAc-T3 can be used to treat disorders associated with hypophosphatemia, such as hypophosphatemic rickets.

Abstract: Aggressive human tumor cells from disparate tissues were found to secrete nucleoside diphosphate kinase-B (NDPK-B) as a phosphoprotein into the extracellular environment. The secreted enzyme was capable of trasphosphorylation activity in the absence of a phosphoryl donor, thereby producing elevated level of extracellular ATP that plays a significant role in angiogenesis required for the growth of cancer cells and cancer metastasis. A series of structurally related non-nucleotide anticancer compounds such as ellagic acid (EA), epigallocatechin gallate (EGCG), and epicatechin gallate (ECG) were found to inhibit the activity of secreted nucleoside diphosphate kinase-B as well as angiogenesis. The nucleoside diphosphate kinase-B inhibition data disclosed herein can be used in predicative models to design novel inhibitors of nucleoside diphosphate kinase-B activity.
Abstract: The present invention provides transgenic mice deficient in corticotropin releasing factor receptor 2 (CRFR2). Mice deficient for CRFR1 exhibit decreased anxiety-like behavior and a decreased stress response. In contrast, CRFR2 null mutant mice are hypersensitive to stress and display increased anxiety-like behavior. These mice are useful for the study of anxiety, depression, and the physiology of the HPA axis. CRFR2 null mutant mice also exhibit increased angiogenesis in all tissues examined. Thus, CRFR2 antagonists may be used to stimulate angiogenesis for the treatment of various conditions. In contrast, CRFR2 agonists may be used to inhibit angiogenesis. A combination of urocortin and bFGF was observed to stimulate rapid hair growth.

Abstract: A peri-arterial blood flow booster apparatus for improving blood pressure and flow, to be implanted around a blood vessel of a patient. The booster comprises a pressure applying device (56) comprising at least one balloon (44), placed alongside a portion of the blood vessel and a restrainer (54) for restraining the balloon and providing counter-forces. It further comprises a control console (60) comprising: an inflating unit (66) for rapidly inflating and deflating the balloon, the inflating unit connected to the balloon; sensing means (68) for sensing electrocardiograph signals of the patient; a control unit (72) for controlling the operation of the inflating unit correlating to the electrocardiograph signals detected by the sensing means. When the balloon is inflated the restrainer forces it to compress the portion of the blood vessel preventing backflow and exerting forces on the blood vessel forcing blood within the portion of the blood vessel to flow antegradely.

Abstract: A human urocortin-related peptide with significant sequence homology to the CRF neuropeptide family was identified. A mouse CDNA was isolated from whole brain poly (A+) RNA that encodes a predicted 38 amino acid peptide protein designated herein as urocortin II. Both human URP and mouse Ucn II are structurally related to the other known mammalian family members, CRF and urocortin (Ucn). These peptides are involved in the regulation of the hypothalamic-pituitary-adrenal axis under basal and stress conditions, suggesting a similar role for URP and Ucn IL Synthesized Ucn-II and URP peptide binds with higher affinity to CRF-R2 than to CRF-R1 Ucn II and human URP appear to be involved in the regulation of body temperature and appetite and may play a role in other stress related phenomenon. These findings identify Ucn II and human URP as a new members of the CRF family of neuropeptides, which are expressed centrally and bind to CRF-R2.
