Abstract: Compositions and methods disclosed concern an isogenic population of in vitro human embryonic stem cells comprising a disease form of the Huntingtin gene (HTT) at the endogenous HTT gene locus in the genome of the cell; wherein the disease form of the HTT gene comprises a polyQ repeat of at least 40 glutamines at the N-terminus of the Huntingtin protein (HTT). The cell lines of the disclosure comprise genetically-defined alterations made in the endogenous HTT gene that recapitulate Huntington's Disease in humans. Furthermore, the cell lines have isogenic controls that share a similar genetic background. Differentiating cell lines committed to a neuronal fate and fully differentiated cell lines are also provided and they also display phenotypic abnormalities associated with the length of the polyQ repeat of the HTT gene. These cell lines are used as screening tools in drug discovery and development to identify substances that fully or partially revert these phenotype abnormalities.

Abstract: A framework for joint computation, caching, and request forwarding in data-centric computing-based networks comprises a virtual control plane, which operates on request counters for computations and data, and an actual plane, which handles computation requests, data requests, data objects and computation results in the physical network. A throughput optimal policy, implemented in the virtual plane, provides a basis for adaptive and distributed computation, caching, and request forwarding in the actual plane. The framework provides superior performance in terms of request satisfaction delay as compared with several baseline policies over multiple network topologies.
Abstract: Described herein are methods and compositions for the prevention or treatment of obesity and obesity-related disorders. The methods and compositions are based, inter alia, on the observations that OPN3 is the most highly expressed opsin in the hypothalamus, a key site for the regulation of energy homeostasis. Indeed, OPN3 expression was highest in regions associated with energy homeostasis, namely the paraventricular nucleus and arcuate nucleus of the hypothalamus. OPN3 was shown to interact and form a complex with MC3R and MC4R, and to modulate MC3R- and MC4R-mediated signaling in the hypothalamus. Accordingly, the methods involve the regulation of melanocortin receptors by downregulating opsin 3 (OPN3) protein expression, OPN3 gene expression, and/or OPN3 activation in the hypothalamus.
Abstract: Surface hydroxyl groups on porous and nonporous metal oxides, such as silica gel and alumina, were metalated with catalyst precursors, such as complexes of earth abundant metals (e.g., Fe, Co, Cr, Ni, Cu, Mn and Mg). The metalated metal oxide catalysts provide a versatile family of recyclable and reusable single-site solid catalysts for catalyzing a variety of organic transformations. The catalysts can also be integrated into a flow reactor or a supercritical fluid reactor.

Abstract: A gait management apparatus applies stimulation to a user suffering from a neurological disease (such as Parkinson's Disease) gait dysfunction. Motion sensors are arranged to be worn by a patient, and electrical stimulation electrodes are on the legs for stimulation. A controller receives motion sensing signals, and processes these signals to generate stimulation signals for operation of the electrodes to stimulate limb movement upon detection of a gait abnormality. There may be a user actuator for user actuation of electrical stimulation, and the inputs may be a series of taps. The controller may provide signals to prevent occurrence of freezing of gait when it senses that a patient is walking or has an intention to walk. Also, it may apply stimulation at an intensity level which is insufficient for functional muscle stimulation but sufficiently high to trigger activation of efferent nerves.

Abstract: Certain donor plasmid vectors such as pFastBac™1 and pFastBac™ Dual lack a cis DNA element upstream of the polh translation start codon (ATG) present in wild type (wt) Autographa californica multiple nucleopolyhedrovirus (AcMNPV), and contain a SV40 pA fragment. When a cis DNA element is inserted upstream of the 50 bp polh promoter and SV40 pA was replaced with a AcMNPV polh pA signal in pFastBac™1 and pFastBac™Dual, certain protein expression levels in High Five™ cells using the Bac-to-Bac® system reached that of the wt AcMNPV.
Abstract: A microwave circulator including an integrated circuit having a number of ports and a respective ring segment coupled to each port to allow microwave frequency signals to be transferred between the port and the respective ring segment. The circulator includes multiple respective ring segments arranged to define multiple parallel circulator rings and at least one superconducting tunnel junction interconnecting each pair of adjacent ring segments and/or a plurality of superconducting tunnel junctions interconnecting each pair of adjacent ring segments to form a circulator ring. The ring segments are configured so that when a bias is applied to the tunnel junctions, signals undergo a phase shift as they traverse the tunnel junctions between ring segments, thereby propagating signals to an adjacent port in a propagation direction.

Abstract: The present invention relates to a multivalent live influenza vaccine platform using a recombinant adenovirus. The present invention is a live attenuated vaccine platform using a recombinant virus and it is easy to inoculate because it is infected with the respiratory tract like influenza virus and exhibits a vaccine action and it is a multivalent vaccine which combines two types into one and it is a highly novel vaccine that does not need to mix viruses compared to vaccines using multiple combinations of one vaccine. The present invention is the first vaccine in which a gene obtained by fusion of two influenza antigen genes into one gene is incorporated into a recombinant virus. Instead of using the entire HA gene of influenza, but using a structurally independent HA1 gene, which is about half of the total HA gene, several types of HA genes could be fused into one.
