    Abstract: Wax-based compositions for making barrier layers used in oral treatment devices are thermally stable when formed into a flat sheet or three-dimensional article to a temperature of at least 45° C. and are plastically deformable at room temperature (25° C.). The wax-based compositions include a wax fraction homogeneously blended with a polymer fraction. The wax fraction includes at least one wax and the polymer fraction includes at least one polymer selected such that, when the at least one wax and at least one polymer are homogeneously blended together, they yield a wax-based composition having the desired properties of thermal stability and plastic deformability. Barrier layers in oral treatment devices made from such wax-based compositions are dimensionally stable to a temperature of at least 40° C. without external support and can be plastically deformed in a user's mouth to become at least partially customized to the size and shape of user's unique dentition.

    Abstract: Methods of identifying target binding molecules by target guided synthesis are provided. The methods include providing two or more fragments capable of reacting to form the target binding molecule and mixing the fragments with the target. The methods can be used to identify target binding molecules that bind targets such as proteins or nucleic acids, including those that bind shallow binding pockets on the surface of such targets. The methods are applied to the Bcl-XL and Mcl-1 proteins from the Bcl-2 family of proteins. Using thio acid and sulfonyl azide fragments capable of reacting through sulfo-click chemistry, new acyl sulfonamides are identified that bind one or both of the Bcl-XL and Mcl-1 proteins. Pharmaceutical formulations of these target binding molecules are also provided.

    Abstract: A control system and method for an engine including a turbocharger without a surge valve involve utilizing one or more pressure sensors configured to measure air pressure in an intake system of the engine, the intake system comprising a compressor of the turbocharger and a throttle valve downstream from the compressor, and a controller configured to control the engine to avoid surge at the compressor and eliminate a need for the surge valve by determining a desired position for the throttle valve based on a driver requested engine torque, determining a minimum mass flow through the compressor that avoids surge based on the measured air pressure in the intake system and a predetermined compressor map, and commanding the throttle valve to a target position to maintain at least the minimum compressor mass flow, wherein the target position is greater than the desired position, thereby avoiding surge at the compressor.
