Abstract: Described is a dispensing needle for a fraction collector. The dispensing needle includes a conduit having a fluid channel to conduct a chromatographic flow, an interior wall that defines the fluid channel, an exterior surface and an endface through which the chromatographic flow is dispensed. The dispensing needle also includes a coating of a hydrocarbon material or a fluorocarbon material that is bonded to the endface. The coating is also bonded to at least a portion of the exterior surface that is adjacent to the endface and at least a portion of the interior wall that is adjacent to the endface. The coating operates to reduce a droplet volume of a liquid dispensed from the endface that may remain at the tip of the dispensing needle. Consequently, the concentration variation in a collected fraction due to a missing droplet or extra droplet is reduced.

Abstract: A method in which a concentration of a chemical species of interest is obtained. The method comprises measuring a property (e.g. the oxidation reduction potential) of a reagent (typically based on a simple single electron redox couple) to obtain a baseline measurement. The reagent is mixed with the solution under test, then the property of the mixture is measured to obtain a post reaction measurement. Then the concentration of the chemical species of interest is determined based on the baseline measurement and the first post reaction measurement, typically by calculating a difference of the baseline measurement and the post reaction measurement, then using the difference and a pre-determined conversion table to determine the concentration of the chemical species of interest.

Abstract: The present inventors show that cannibal cells can undergo senescence after entosis in vivo and that the tumor suppressive protein p53 act as a repressor of this phenomenon. They therefore propose new tools to study the molecular pathways involved in the cannibalism process, for example by measuring the expression levels of p53 or splice variants thereof (such as ?133TP53, TP53?, TP53? or ?40TP53), the release of extracellular ATP or purinergic P2Y2 receptor activity. The present inventors also demonstrated that the detection of senescent cannibal cells in breast adenocarcinoma obtained from patients treated with neo-adjuvant therapy positively correlates with good patient's response to treatment. Altogether, these results provide the first evidence that detection of cellular cannibalism and senescence simultaneously in tumors helps for the diagnosis of disease outcomes and for the prediction of treatment efficiency against cancer diseases.

Abstract: Cancer patients make antibodies to tumor-derived proteins that are potential biomarkers for early detection. Twenty-eight antigens have been identified as potential biomarkers for the early detection of basal-like breast cancer (Tables 1, 2). Also, a 13-AAb classifier has been developed that differentiate patients with BLBC from healthy controls with 33% sensitivity at 98% specificity (Table 3).

Abstract: Provided herein are kits for detecting and quantifying the level of a metastatic cancer biomarker(s) in a patient. The kits may contain a detection and quantifying means, such as, biomarker specific antibodies. Also provided are methods for determining a prognosis, a stratification and/or monitoring of a metastatic cancer in a patient. The level or activity of at least two biomarkers, such as obscurin, PI3K, E-cadherin, or N-cadherin is measured in a patient sample and compared to corresponding biomarkers in a control sample. A significant difference in the level or activity of the biomarkers between the patient sample and control is indicative of a metastatic cancer. In addition a method for treating a metastatic breast cancer via administration of a PI3K inhibitor and an agent that increases expression of obscurin is provided.

Abstract: The present invention provides an approach for the determination of the activation states of a plurality of proteins in single cells. This approach permits the rapid detection of heterogeneity in a complex cell population based on activation states, expression markers and other criteria, and the identification of cellular subsets that exhibit correlated changes in activation within the cell population. Moreover, this approach allows the correlation of cellular activities or properties. In addition, the use of modulators of cellular activation allows for characterization of pathways and cell populations.

Abstract: A method for analyzing a sample includes trapping a first ion packet in a first segment of a multi-segment reaction cell; trapping a second ion packet in a second segment of the multi-segment reaction cell; and trapping a third ion packet in a third segment of the multi-segment reaction cell. At least one of the first, second, and third ion packets includes precursor ions, and at least another one of the first, second, and third ion packets includes reagent ions. The method further includes mixing the first, second, and third ion packets within the reaction cell to cause a reaction between the precursor ions and the reagent ions to form product ions.
