Abstract: The presence of mycotoxins in agricultural products necessitates large scale testing of a wide range of sample material to ensure the safety of food and feed. The mycotoxin ochratoxin A represents an enablement for all mycotoxins as the level of sensitivity necessary for regulatory requirements for this compound at the part per billion level are as low or lower than any other mycotoxin. This invention describes the identification of a set of DNA ligands with sufficiently high binding affinity and specificity for ochratoxin A to enable an improvement over existing methods for the separation, concentration and quantitative determination of ochratoxin A in sample material.

Abstract: The present invention relates to the creation of broad-spectrum resistance in transgenic plants by inserting inhibitory nucleic acid sequences inhibiting the expression of fungal genes. The nucleic acid sequences of genes which are crucial to development, growth and profilation of fungi, i.e. sequences of genes with an essential function in fungi, are often conserved and/or have a high sequence identity. Regions having a particularly high sequence identity between various fungi are used in order to produce inhibitory gene constructs, e.g. on basis of antisense, siRNA, shRNA, ribozyme technology and other technologies imparting the inhibition of the expression and activation of genes. Since said methods base on the sequence-specific hybridization of the inhibitory RNA molecules with respective target sequences in certain fungal genes, the expression of all corresponding genes from various fungi is thus inhibited as well.

Abstract: The present invention describes a novel molecular method based on the polymerase chain reaction (PCR) in a multiplex variant in order to detect and identify Candida species with clinical relevance, namely C. albicans, C. glabrata, C. krusei, C. parapsilosis, C. tropicalis, C. guilliermondii, C. lusitaniae e C. dubliniensis. The strategy uses the existence of sequences, whether conserved or variable, in fungal ribosomal genes and in the use of a combination of universal primers, specific for fungi, and internal primers, specific for each one of the Candida species (FIG. 1). In this sense, two fragments from the internal transcribed spacer (ITS) regions of the ribosomal RNA (rRNA) are amplified by multiplex PCR, allowing the easy identification of the Candida species in question. This methodology allows a rapid, effective and low-cost identification of Candida species with clinical relevance, bearing several advantages over the currently available diagnostic methods.

Abstract: Compositions and methods for protecting a plant from a pathogen, particularly a fungal pathogen, are provided. Compositions include novel amino acid sequences, and variants and fragments thereof, for antipathogenic polypeptides that were isolated from microbial fermentation broths. Nucleic acid molecules comprising nucleotide sequences that encode the antipathogenic polypeptides of the invention are also provided. A method for inducing pathogen resistance in a plant using the nucleotide sequences disclosed herein is further provided. The method comprises introducing into a plant an expression cassette comprising a promoter operably linked to a nucleotide sequence that encodes an antipathogenic polypeptide of the invention. Compositions comprising an antipathogenic polypeptide or a transformed microorganism comprising a nucleic acid of the invention in combination with a carrier and methods of using these compositions to protect a plant from a pathogen are further provided.

Abstract: The invention is generally related to methods of generating plants transformed with novel autonomous mini-chromosomes. Mini-chromosomes with novel compositions and structures are used to transform plants cells which are in turn used to generate the plant. Methods for generating the plant include methods for delivering the mini-chromosome into plant cell to transform the cell, methods for selecting the transformed cell, and methods for isolating plants transformed with the mini-chromosome. Plants generated in the present invention contain novel genes introduced into their genome by integration into existing chromosomes.
