    Abstract: A process to obtain linear double-stranded covalently closed DNA “dumbbell” constructs from plasmids by restriction digest, subsequent ligation with hairpin oligodesoxyribonucleotides, optionally in the presence of restriction enzyme, and a final digestion with endo- and exonucleolytic enzymes that degrade all contaminating polymeric DNA molecules but the desired construct. The invention also provides a process to obtain said dumbbell constructs employing endonuclease class II enzymes. Furthermore, the invention provides a process to obtain linear, covalently closed DNA molecules, such as plasmids, free from contamination by genomic DNA, by submitting the DNA preparation to a facultative endonucleolytic degradation step and an obligatory exonucleolytic degradation step.

    Abstract: A door lock for an all-glass door with an adjacent fixed glass side panel. Furthermore, the door lock comprises a first lock casing arranged on one side of the all-glass door and a second lock casing placed opposite to the first lock casing on the other side of the all-glass door. The first lock casing comprises the latch bolt and the second lock casing comprises the strike plate for the dead bolt. Furthermore, there are a first lock strike box and a second lock strike box, effectively connected to each other by means of a closing cylinder through a boring contained in the fixed glass side panel.

The arc is established by lifting the welding torch (12) after contact between electrode (13) and workpiece (14) under low ignition current. When the arc length is sufficient, the welder orders a change-over to welding current via an operating device (11). The invention can also be used for controlling the supply of heat to the workpiece both during continuous bead application and during the crater filling time at the end of welding. The invention creates reliable ignitions without extensive use of control electronics, and it substantially reduces the electrode wear in manual TIG welding.
