Abstract: An osmotic device is disclosed for delivering a beneficial agent. The device comprises a wall surrounding a compartment containing an agent, a passageway in the wall connecting the exterior of the device with the compartment, and (a) an agent in the wall or (b) an agent carried on the wall in the wall. The device delivers the agent from the compartment and the wall.

Abstract: A method for transdermal delivery of naloxone, naltrexone and nalbuphine base through intact skin is described. Preferred embodiments of transdermal therapeutic systems for delivering these drugs and polyethylene glycol monolaurate employ an ethylene vinylacetate matrix containing drug base at a concentration above saturation and polyethylene glycol monolaurate below unit activity. Polyethylene glycol manolaurate is disclosed as a permeation enhancer for the base form of these drugs and is preferably delivered simultaneously with the drug.

Abstract: The release pattern of a dispersion type active agent dispenser can be improved by forming the device from a depleted zone containing the agent at a concentration no greater than the saturation and a non-depleted zone containing the agent dispersed in the matrix at a uniform concentration greater than saturation. The depleted zone is disposed between the surface through which the agent is to be dispensed and at least a substantial portion of the non-depleted zone with the interface therebetween disposed at a non-uniform distance from the releasing surface. Certain embodiments are particularly useful for the transdermal administration of drugs.

Abstract: Mineral oil (MO) polyisobutylene (PIB), colloidal silicon dioxide (CSD) mixtures suitable for use as drug containing matrices in transdermal delivery systems are disclosed. Preferred systems for dispensing moderately mineral oil soluble drugs contain at least about 6% CSD, have a MO/PIB of at least 1.0 and a viscosity of at least 1.5.times.10.sup.7 poises. Preferred systems for dispensing clonidine have a clonidine permeability of at least 1.0.times.10.sup.-4 .mu.g/cm sec and a MO/PIB of at least 1.2.
Abstract: An osmotic delivery system is disclosed for dispensing a useful agent. The system comprises a semipermeable wall surrounding a compartment housing the agent. The wall comprises a cellulose ether, an organic solvent soluble polymer, and optionally an aqueous soluble polymer. A passageway in the wall connects the interior of the system with the exterior for dispensing an agent from the system.
Abstract: A self-contained body mounted pump assembly for continuously administering a therapeutic agent parenterally. The pump has a transparent top through which the contents can be seen and outwardly flaring sides to provide for comfort, stability and other useful features. The pump assembly is driven by a fluid imbibing, preferably osmotic pump and contains its own source of actuating fluid. Means for superimposing a pulse dose on the tonic flow are also described.

The active agent is dispensed from the pump after the pump is exposed to the fluid. Means to vary or stop the pump by obdurating all or a portion of the exterior surface of the pump can also be provided. Variation in flow rates can also be obtained by changing the composition of the actuating fluid.

Abstract: A formulation dispenser is disclosed for use in a parenteral delivery system. The dispenser comprises an inlet for admitting fluid into the dispenser and an outlet for letting fluid leave the dispenser. A beneficial agent is housed in a formulation chamber in the dispenser and a membrane permeable to fluid and agent defines one boundary of the formulation chamber. A piston is slideably disposed in the dispenser for governing the flow of fluid through the dispenser, or for permitting fluid to contact the membrane and enter the formulation chamber for forming a solution containing agent that flows through the outlet into the parenteral system.
