    Abstract: The present invention provides a protocol and apparatus for enriching circulating tumor cells and other rare cells from blood, including debris and other components, from samples with high precision and at high throughput rates. This invention discloses an improved processing system from previously described semi-automated sample processing. The system further reduces operator intervention and hands-on time from prior systems. While this system has general utility in processing diverse materials, the system is configured for sample processing of biological specimens to provide an enriched fraction suitable for detection, enumeration and identification of target cells by appropriate analytical methodologies.

The system also includes a cloner to replicate the elite sets, and a sampler to modify the elite sets so that each elite set is mutually statistically independent while still yielding query-result values above the cutoff value.

When the number of attempts is not greater than the threshold, the method includes sending a service unavailable message to the client device. A retry-after delay may be calculated by the server according to a predicted time for the server to exit the overloaded state and included in the service unavailable message. The predicted time may take into account both requests currently being serviced and requests that have been scheduled to be serviced in the future.

    Abstract: The invention in one variation is a pressure vessel having a pair of hemispheres with a wall and a circular edge, a pair of sealing elements, and an intervening mounting plate with circular groove on both sides of the plate. The hemispheres are mounted on opposite sides of the plate. Each groove has an sealing element and the circular edge. A seal is formed between the plate and a hemisphere when the hemisphere is pressed against the sealing element. Pressure can be generated by creating a partial vacuum in the sealed hemispheres or by using clamps. When exposed to higher pressures, such as when the vessel is used underwater, the pressure on the hemispheres is conveyed to the sealing element forming an even tighter seal. The vessel has interface connections that enable communication to and from devices mounted inside. The vessels are modular and can be ganged to provide redundancy or different functions.
