Abstract: Polymeric thin-film tube connectors, bioreactors, systems and methods are described, the connectors allowing connection of a polymeric thin-film tube of a bioreactor chamber with a complementary part, typically, rigid port of flange of the bioreactor or bioreactor system. Further, bioreactors having a thin-film reactor chamber and one or more connectors are described. The thin-film reactor chamber can have a thin-film wall for enclosing culture medium and microorganisms. The connector can include a flexible boot. An opening of the thin-film wall couples to the flexible boot. The thin-film wall can extend through a reactor end of the flexible boot and can rest against an interior surface of the flexible boot. The connectors are particularly advantageous for connecting standard utilities in a bioreactor system to thin-film bioreactor chambers or capsules having a thin-film tubular opening.

  • Publication number: 20140234967
Abstract: A method to generate siRNAs in vivo is described, as are constructs and compositions useful in the method. The method does not depend on the use of DNA or synthetic constructs that contain inverted duplications or dual promoters so as to form perfect or largely double-stranded RNA. Rather, the method depends on constructs that yield single-stranded RNA transcripts, and exploits endogenous or in vivo-produced miRNAs or siRNAs to initiate production of siRNAs. The miRNAs or siRNAs guide cleavage of the transcript and set the register for production of siRNAs (usually 21 nucleotides in length) encoded adjacent to the initiation cleavage site within the construct. The method results in specific formation of siRNAs of predictable size and register (phase) relative to the initiation cleavage site. The method can be used to produce specific siRNAs in vivo for inactivation or suppression of one or more target genes or other entities, such as pathogens.

  • Publication number: 20130137178
Abstract: The present invention relates to a cell line derived from the cambium of an herbaceous plant having a storage root and a method for isolating the same. More specifically, relates to a cambium-derived homogeneous cell line having the ability to divide, which is obtained from the cambium-containing storage tissue of an herbaceous plant having a storage root without a separate dedifferentiation process, and to a method for isolating the same. The cell line derived from the cambium of an herbaceous plant having a storage root has an active division ability and is homogeneous. Also, it is stable during culture, because it has not undergone a dedifferentiation process. Thus, through the optimization of proliferation thereof, the cell line can be allowed to proliferate in a large amount within a short time.

Abstract: A fluid mixing system includes a support housing having an interior surface bounding a chamber. A flexible bag is disposed within the chamber of the support housing, the flexible bag having an interior surface bounding a compartment. An impeller is disposed within the chamber of the flexible bag. A drive shaft is coupled with the impeller such that rotation of the drive shaft facilitates rotation of the impeller. A drive motor assembly is coupled with the draft shaft and is adapted to rotate the drive shaft. An adjustable arm assembly is coupled with the drive motor assembly and is adapted to move the drive motor assembly which in turn moves the position of the drive shaft and impeller. An electrical controller can control movement of the adjustable arm.
