Abstract: A non-naturally occurring microorganism having a 1,3-BDO pathway is provided. The microorganism expresses at least one of the following 1,3-BDO pathway enzymes: an aldolase that catalyzes condensation of two acetaldehydes to produce 3-hydroxybutanal; and an aldo-ketoreductase, oxidoreductase, aldehyde reductase or alcohol dehydrogenase that reduces 3-hydroxybutanal to 1,3-BDO. The organism may further express one or more enzymes for producing acetaldehyde. A biosynthetic process involves condensing two acetaldehyde molecules to 3-hydroxybutanal using an enzyme from class aldolases; and selectively reducing 3-hydroxybutanal to 1,3-BDO using an enzyme belonging to the class aldo-ketoreductase, oxidoreductase, aldehyde reductase or alcohol dehydrogenase. • Patent number: 9593303
Abstract: The present invention relates to a method of culturing bacterial organisms belonging to the family Anaplasmataceae in mammalian embryonic or fetal cells. In particular, the present invention is directed to growth of bacterial organisms belonging to the family Anaplasmataceae including organisms belonging to the Anaplasma, Ehrlichia and Neorickettsia genera. The bacterial organisms may be cultured in mammalian embryonic or fetal host cells including feline embryonic host cells. Bacterial material cultured according to the methods described herein may be used as the basis for vaccines against diseases associated with the Anaplasmataceae bacteria, or as the basis for diagnostic applications useful for diagnosing diseases associated with the Anaplasmataceae bacteria.

  • Patent number: 9562260
Abstract: An instrument for conducting nucleic acid amplification reactions in a disposable test device. The test device includes a first reaction chamber containing a first nucleic acid amplification reagent (e.g., primers and nucleotides) and a second reaction chamber either containing, or in fluid communication, with a second nucleic acid amplification reagent (e.g., an amplification enzyme such as RT). The instrument includes a support structure receiving the test device. A temperature control system maintains the first reaction chamber at a first elevated temperature but simultaneously maintains the second nucleic acid amplification reagent at a second temperature lower than the first temperature so as to preserve the second nucleic acid amplification reagent. An actuator operates on a fluid conduit in the test device to place the first and second reaction chambers in fluid communication with each other after a reaction has occurred in the first reaction chamber at the first temperature.
